A goal in synthetic biology is to modify cells so that they synthesize artificial amino acids and string them together with natural amino acids to form proteins that no other form of life can make.

In 1989 Steven Benner led a team at ETH Zurich that created DNA containing two artificial genetic “letters”. No one had made genes from altered DNA that were functional.

In 2004, scientists at the Scripps Research Institute produced six kinds of unnatural amino acids in yeast and began adapting them to work in Human cells and Roundworms. [The Arthropod Connection]

The translational machinery that reads RNA to make proteins in Yeast is very similar to the translational machinery of humans.

This system can make two different synthetic amino acids and put them in the same protein. The cell decodes a 4-nucleotide sequence with which we can attach polymers to proteins.

The two variants, TNA in which ribose is replaced with a simpler sugar & x DNA are more stable than DNA for reprogramming cells. First, we have to get them working inside living organisms.
In the past, using proteins in large quantities has been limited because proteins must exist in an environment which mimics that found in a cell.

What is being done with the new biomaterials?
In molecular fabrication, chemical and structure must work together with the water-mediated hydrogen bond of living systems.

What Are Quantum Dots?
Quantum dots are artificial atoms. They are clouds of electrons with nucleus (protons and neutrons) trapped in a very small crystal structure by electronic components.
In a real atom, electron clouds exist in multiple layers above the atomic nucleus. In quantum dots, the electrons are trapped in a space so small that their wave-like behaviour dominates over their particle-like behaviour.
When an electron jumps to a higher layer, it absorbs energy (in the form of heat, light, mechanical vibration), and when it falls to a lower level, it emits energy (usually in the form of light).
Quantum dots obey quantum-mechanical rule called the Pauli Exclusion Principle, acting like real atoms by arranging themselves in patterns.
Quantum dots emit a particular wavelength and colour depending on their composition and size. Their size is controlled by adding or subtracting electrons.

Can quantum dots penetrate human skin?
Skin is the largest organ of the body and is a potential route of exposure to nanomaterials. Quantum dots of different sizes, shapes, and surface coatings can penetrate the intact skin at a relevant dose within 8-24 hrs.

Peptides enhance skin penetration when conjugated to drug molecules and quantum dots and can target specific tissues. Peptides are short pieces of amino acids.

Biodistribution studies revealed that particle clearance from the lungs and extrapulmonary distribution was, in part, size dependent. Microparticles dispersed but nanoparticles stayed in the lung.
Microparticles were rapidly dispersed into the body by mucus and through blood circulation.
Nanoparticles enhanced by Arg9 were restricted to the lung but did not induce inflammation.
